Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.943C>T (p.His315Tyr), citing Ambry Variant Classification Scheme 2023: The p.H315Y variant (also known as c.943C>T), located in coding exon 11 of the MLH1 gene, results from a C to T substitution at nucleotide position 943. The histidine at codon 315 is replaced by tyrosine, an amino acid with similar properties. In one study, this variant was detected in 0/165 colorectal cancer and/or polyposis patients and was identified in 1/2512 control individuals from a healthy population database (Rosenthal EA et al. Hum Genet, 2018 Oct;137:795-806). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
