NM_000249.4(MLH1):c.943C>T (p.His315Tyr) was classified as Uncertain Significance for Lynch syndrome by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This missense variant replaces histidine with tyrosine at codon 315 of the MLH1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a general population control and absent in colorectal cancer or polyps cases (PMID: 30267214). This variant has been identified in 3/251402 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
