Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.672C>A (p.Ser224Arg), citing Ambry Variant Classification Scheme 2023: The p.S224R variant (also known as c.672C>A), located in coding exon 2 of the HOXB13 gene, results from a C to A substitution at nucleotide position 672. The serine at codon 224 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved through mammals, but not in lower available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
