NM_006361.6(HOXB13):c.728A>G (p.Lys243Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K243R variant (also known as c.728A>G), located in coding exon 2 of the HOXB13 gene, results from an A to G substitution at nucleotide position 728. The lysine at codon 243 is replaced by arginine, an amino acid with highly similar properties. This variant was identified in a cohort of Iranian prostate cancer patients (Roudi R et al. Med J Islam Repub Iran, 2018 Oct;32:97). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
