Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.137T>C (p.Leu46Pro), citing Ambry Variant Classification Scheme 2023: The p.L46P variant (also known as c.137T>C), located in coding exon 2 of the CDH1 gene, results from a T to C substitution at nucleotide position 137. The leucine at codon 46 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
