Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.8745G>C (p.Gln2915His), citing Ambry Variant Classification Scheme 2023: The c.8745G>C (p.Q2915H) alteration is located in exon 52 (coding exon 52) of the DNAH5 gene. This alteration results from a G to C substitution at nucleotide position 8745, causing the glutamine (Q) at amino acid position 2915 to be replaced by a histidine (H). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
