NM_000059.4(BRCA2):c.665C>T (p.Pro222Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P222L variant (also known as c.665C>T), located in coding exon 7 of the BRCA2 gene, results from a C to T substitution at nucleotide position 665. The proline at codon 222 is replaced by leucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
