NM_007294.4(BRCA1):c.4568C>T (p.Pro1523Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P1523L variant (also known as c.4568C>T), located in coding exon 13 of the BRCA1 gene, results from a C to T substitution at nucleotide position 4568. The proline at codon 1523 is replaced by leucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
