NM_000465.4(BARD1):c.1847G>A (p.Ser616Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This missense variant replaces serine with asparagine at codon 616 of the BARD1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). A functional study has shown that mammalian cells expressing this variant protein exhibit comparable homology directed DNA repair activity to cells expressing the wild-type protein in an in vitro assay (PMID: 30925164). To our knowledge, this variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/251116 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
