Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002382.5(MAX):c.404A>G (p.Asp135Gly), citing Ambry Variant Classification Scheme 2023: The p.D135G variant (also known as c.404A>G), located in coding exon 5 of the MAX gene, results from an A to G substitution at nucleotide position 404. The aspartic acid at codon 135 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
