Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.1395T>G (p.Cys465Trp), citing Ambry Variant Classification Scheme 2023: The p.C465W variant (also known as c.1395T>G), located in coding exon 9 of the ATM gene, results from a T to G substitution at nucleotide position 1395. The cysteine at codon 465 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
