Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000245.4(MET):c.2888-1G>C, citing Ambry Variant Classification Scheme 2023: The c.2942-1G>C intronic variant results from a G to C substitution one nucleotide upstream from coding exon 13 of the MET gene. Variants that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this variant remains unclear.
