Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.1335G>C (p.Gln445His), citing Ambry Variant Classification Scheme 2023: The p.Q445H variant (also known as c.1335G>C), located in coding exon 10 of the APC gene, results from a G to C substitution at nucleotide position 1335. The glutamine at codon 445 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this variant remains unclear.
