NM_177438.3(DICER1):c.2144T>A (p.Val715Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V715D variant (also known as c.2144T>A), located in coding exon 13 of the DICER1 gene, results from a T to A substitution at nucleotide position 2144. The valine at codon 715 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
