Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.2136G>T (p.Glu712Asp), citing Ambry Variant Classification Scheme 2023: The p.E712D variant (also known as c.2136G>T), located in coding exon 23 of the FANCA gene, results from a G to T substitution at nucleotide position 2136. The glutamic acid at codon 712 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
