Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.2500A>G (p.Lys834Glu), citing Ambry Variant Classification Scheme 2023: The p.K834E variant (also known as c.2500A>G), located in coding exon 4 of the TNXB gene, results from an A to G substitution at nucleotide position 2500. The lysine at codon 834 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
