Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000784.4(CYP27A1):c.800T>G (p.Phe267Cys), citing Ambry Variant Classification Scheme 2023: The p.F267C variant (also known as c.800T>G), located in coding exon 4 of the CYP27A1 gene, results from a T to G substitution at nucleotide position 800. The phenylalanine at codon 267 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
