Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.2189A>G (p.Glu730Gly), citing Ambry Variant Classification Scheme 2023: The p.E730G variant (also known as c.2189A>G), located in coding exon 9 of the BRCA1 gene, results from an A to G substitution at nucleotide position 2189. The glutamic acid at codon 730 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
