Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.194G>C (p.Cys65Ser), citing Ambry Variant Classification Scheme 2023: The p.C65S variant (also known as c.194G>C), located in coding exon 2 of the TSC2 gene, results from a G to C substitution at nucleotide position 194. The cysteine at codon 65 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
