Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.18410G>A (p.Gly6137Asp), citing Ambry Variant Classification Scheme 2023: The p.G5180D variant (also known as c.15539G>A), located in coding exon 56 of the OBSCN gene, results from a G to A substitution at nucleotide position 15539. The glycine at codon 5180 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
