Uncertain significance — the classification assigned by Ambry Genetics to NM_130384.3(ATRIP):c.2189T>C (p.Leu730Pro), citing Ambry Variant Classification Scheme 2023: The p.L730P variant (also known as c.2189T>C), located in coding exon 12 of the ATRIP gene, results from a T to C substitution at nucleotide position 2189. The leucine at codon 730 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
