NM_003114.5(SPAG1):c.1621T>G (p.Tyr541Asp) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y541D variant (also known as c.1621T>G), located in coding exon 12 of the SPAG1 gene, results from a T to G substitution at nucleotide position 1621. The tyrosine at codon 541 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
