This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 5880, where C is replaced by G; at the protein level this means replaces isoleucine at residue 1960 with methionine — a missense variant. Submitter rationale: The p.I1960M variant (also known as c.5880C>G), located in coding exon 38 of the ATM gene, results from a C to G substitution at nucleotide position 5880. The isoleucine at codon 1960 is replaced by methionine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6498 samples (12996 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 125000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
