Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020207.7(ERCC6L2):c.1853A>G (p.Tyr618Cys), citing Ambry Variant Classification Scheme 2023: The p.Y618C variant (also known as c.1853A>G), located in coding exon 13 of the ERCC6L2 gene, results from an A to G substitution at nucleotide position 1853. The tyrosine at codon 618 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
