Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.6512_6519dup (p.Ser2174fs), citing Ambry Variant Classification Scheme 2023: The c.6512_6519dupCCACACTT pathogenic mutation, located in coding exon 44 of the ATM gene, results from a duplication of CCACACTT at nucleotide position 6512, causing a translational frameshift with a predicted alternate stop codon (p.S2174Pfs*64). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
