The phenylalanine at codon 153 is replaced by serine, an amino acid with highly dissimilar properties. This variant was reported in individual(s) with features consistent with familial hypercholesterolemia (FH) (Hori M et al. Atherosclerosis, 2019 Oct;289:101-108; Tada H et al. J Clin Lipidol, 2020 Mar;14:346-351.e9; Kim H et al. J Atheroscler Thromb, 2022 Aug;29:1176-1187). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
