NM_002382.5(MAX):c.427G>C (p.Glu143Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E143Q variant (also known as c.427G>C), located in coding exon 5 of the MAX gene, results from a G to C substitution at nucleotide position 427. The glutamic acid at codon 143 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
