NM_000245.4(MET):c.19C>T (p.Leu7Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L7F variant (also known as c.19C>T), located in coding exon 1 of the MET gene, results from a C to T substitution at nucleotide position 19. The leucine at codon 7 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
