Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.1039G>C (p.Ala347Pro), citing Ambry Variant Classification Scheme 2023: The p.A347P variant (also known as c.1039G>C), located in coding exon 8 of the CDH1 gene, results from a G to C substitution at nucleotide position 1039. The alanine at codon 347 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
