This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 553, where C is replaced by G; at the protein level this means replaces histidine at residue 185 with aspartic acid — a missense variant. Submitter rationale: The p.H185D variant (also known as c.553C>G), located in coding exon 6 of the PTEN gene, results from a C to G substitution at nucleotide position 553. The histidine at codon 185 is replaced by aspartic acid, an amino acid with similar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally deficient (Mighell TL et al.
