Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.1835C>T (p.Ser612Leu), citing Ambry Variant Classification Scheme 2023: The p.S612L variant (also known as c.1835C>T), located in coding exon 12 of the FLNA gene, results from a C to T substitution at nucleotide position 1835. The serine at codon 612 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
