Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_017654.4(SAMD9):c.869T>G (p.Val290Gly), citing Ambry Variant Classification Scheme 2023: The p.V290G variant (also known as c.869T>G), located in coding exon 1 of the SAMD9 gene, results from a T to G substitution at nucleotide position 869. The valine at codon 290 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
