Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.357delinsCTCTTAAATTTGCATATGTTATAAATAATGTCAATATAAAAACTGATAGCATGG (p.Asp119_Val120insSerTer), citing Ambry Variant Classification Scheme 2023: The c.357delTins54 pathogenic mutation, located in coding exon 5 of the PMS2 gene, results from the deletion of one nucleotide and insertion of 54 nucleotides (CTCTTAAATTTGCATATGTTATAAATAATGTCAATATAAAAACTGATAGCATGG) causing a translational frameshift with a predicted alternate stop codon (p.V120Sfs*2). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
