Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.6298C>G (p.Gln2100Glu), citing Ambry Variant Classification Scheme 2023: The p.Q2100E variant (also known as c.6298C>G), located in coding exon 10 of the BRCA2 gene, results from a C to G substitution at nucleotide position 6298. The glutamine at codon 2100 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
