NM_000038.6(APC):c.4282G>A (p.Gly1428Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1428R variant (also known as c.4282G>A), located in coding exon 15 of the APC gene, results from a G to A substitution at nucleotide position 4282. The glycine at codon 1428 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
