NM_058216.3(RAD51C):c.28A>T (p.Met10Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M10L variant (also known as c.28A>T), located in coding exon 1 of the RAD51C gene, results from an A to T substitution at nucleotide position 28. The methionine at codon 10 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
