Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.1897A>G (p.Lys633Glu), citing Ambry Variant Classification Scheme 2023: The p.K633E variant (also known as c.1897A>G), located in coding exon 5 of the PALB2 gene, results from an A to G substitution at nucleotide position 1897. The lysine at codon 633 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
