This variant was reported in individual(s) with features consistent with neurofibromatosis type 1; in at least one individual, it was determined to be de novo (Koczkowska M et al. Hum Genet, 2023 Jul;142:849-861; Ambry internal data). This nucleotide position is conserved on limited sequence alignment. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice donor site.
