NM_005826.5(HNRNPR):c.311C>T (p.Thr104Ile) was classified as Uncertain Significance for Neurodevelopmental disorder by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard, citing ACMG Guidelines, 2015: The heterozygous p.Thr104Ile variant in HNRNPR was identified in 1 individual with a neurodevelopmental disorder including autism via a collaborative study between the Broad Institute's Center for Mendelian Genomics and the NeuroDev Study (https://www.neurodevproject.org/). Trio exome analysis showed this variant to be de novo. The p.Thr104Ile variant in HNRNPR has not been previously reported in the literature in individuals with neurodevelopmental disorders, and was absent from large population studies. Computational prediction tools and conservation analyses do not provide strong support for or against an impact to the protein. The number of missense variants reported in HNRNPR in the general population is lower than expected, suggesting there is little benign variation in this gene and slightly increasing the possibility that a missense variant in this gene may not be tolerated.
