This is a single-copy gain (three copies) of the chr5:175416095-177250474 region (~1.83 Mb) on cytogenetic band 5q35.2-35.3. Submitter rationale: A 1.8 Mb duplication at 5q35.2–q35.3 encompassing NSD1 was identified by chromosomal microarray analysis. The duplication occurred de novo. This region includes NSD1, which has a ClinGen triplosensitivity score of 3, indicating sufficient evidence for dosage pathogenicity. The variant is consistent with previously reported pathogenic 5q35 microduplications.
