Submitter rationale: We identified a novel homozygous single-nucleotide deletion within exon 6 of the DENND5A gene (NM_015213.4: c.1266delT). This frameshift variant is predicted to result in nonsense-mediated mRNA decay (NMD). The novel DENND5A variant was validated using Sanger sequencing. Based on the ACMG/AMP standards and guidelines, it was classified as pathogenic based on the following criteria: PVS1 (Very Strong), PM2 (Moderate), PM3 (Moderate).
