Pathogenic for Partial deletion of the long arm of chromosome 7 — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to Single allele, citing ACMG/ClinGen CNV Guidelines, 2019: A confirmed de novo heterozygous deletion of 7q21.2-q22.1. ([GRCh38] chr7:91872849_100457083x1) encompassing 89 genes (https://genescout.omim.org/) was identified by exome sequencing of one individual with hearing abnormality, mild global developmental delay, delayed fine motor development, delayed gross motor development, abnormal social behavior, and delayed speech and language development, via a collaborative study between the Broad Institute's Center for Mendelian Genomics and the NeuroDev Study (https://www.neurodevproject.org/). These breakpoints have been called by exome sequencing only and therefore may not reflect the true breakpoints. The patient phenotype is nonspecific, but is consistent with cases described in the literature and/or published databases with overlapping variants. There is complete overlap with the SGCE gene which is known to be haploinsufficient and has been assessed by the ClinGen Dosage Sensitivity Working Group (https://search.clinicalgenome.org/kb/gene-dosage). Data from large population studies are insufficient to assess the frequency of this variant. In summary, this variant meets criteria to be classified as pathogenic for 7q deletion syndrome.
