Uncertain significance for Primary dilated cardiomyopathy; Danon disease — the classification assigned by Clinical Genomics Laboratory, Stanford Medicine to NM_002294.3(LAMP2):c.418C>A (p.Leu140Ile), citing ACMG Guidelines, 2015: The p.Leu140Ile variant in the LAMP2 gene has not been previously reported in association with disease. This variant was absent from large population databases, including the Genome Aggregation Database (http://gnomad.broadinstitute.org/). The leucine at position 140 is poorly evolutionarily conserved. Computational tools predict that the p.Leu140Ile variant does not impact protein function; however, the accuracy of in silico algorithms is limited. These data were assessed using the ACMG/AMP variant interpretation guidelines. In summary, the significance of the p.Leu140Ile variant is uncertain.
