Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.911G>A (p.Cys304Tyr), citing Ambry Variant Classification Scheme 2023. This variant lies in the MUTYH gene (transcript NM_001048174.2) at coding-DNA position 911, where G is replaced by A; at the protein level this means replaces cysteine at residue 304 with tyrosine — a missense variant. Submitter rationale: The p.C332Y variant (also known as c.995G>A), located in coding exon 11 of the MUTYH gene, results from a G to A substitution at nucleotide position 995. The cysteine at codon 332 is replaced by tyrosine, an amino acid with highly dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 200000 alleles tested) in our clinical cohort. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
