Likely pathogenic for Gaucher disease — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000157.4(GBA1):c.599T>C (p.Ile200Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GBA1 c.599T>C (p.Ile200Thr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The frequency of this variant in the general population could not be determined as the technology used for large population databases (ExAC, gnomAD, ESP, 1000G) cannot detect variants of this type. c.599T>C has been observed in the presumed compound heterozygous state in at least 1 and in the homozygous state in at least 1 additional individual(s) affected with Gaucher Disease (example, Thomas_2021, Mozafari_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. A different variant affecting the same codon has been classified as likely pathogenic/pathogenic (c.599T>A, p.Ile200Asn), supporting the critical relevance of codon 200 to GBA1 protein function.
