This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 4810, where G is replaced by A; at the protein level this means replaces glycine at residue 1604 with arginine — a missense variant. Submitter rationale: Variant summary: COL7A1 c.4810G>A (p.Gly1604Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251280 control chromosomes (gnomAD). c.4810G>A has been observed in individuals affected with Dystrophic Epidermolysis Bullosa, Recessive (e.g. Whittock_1999, Venugopal_2013). These data indicate that the variant may be associated with disease. This variant disrupts the triple helix domain of COL7A1. Glycine residues within the Gly-Xaa-Yaa repeats of the triple helix domain are required for the structure and stability of fibrillar collagens (PMID: 7695699, 8218237, 19344236). A different variant affecting the same codon has been classified as pathogenic by our lab (c.4811G>A, p.Gly1604Glu), supporting the critical relevance of codon 1604 to COL7A1 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
