Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.10G>A (p.Glu4Lys), citing Ambry Variant Classification Scheme 2023: The p.E4K variant (also known as c.10G>A), located in coding exon 1 of the CHEK2 gene, results from a G to A substitution at nucleotide position 10. The glutamic acid at codon 4 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
