Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.629C>G (p.Ser210Ter), citing Ambry Variant Classification Scheme 2023: The p.S210* pathogenic mutation (also known as c.629C>G), located in coding exon 4 of the CHEK2 gene, results from a C to G substitution at nucleotide position 629. This changes the amino acid from a serine to a stop codon within coding exon 4. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
