Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004360.5(CDH1):c.1843A>G (p.Ile615Val), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the CDH1 gene (transcript NM_004360.5) at coding-DNA position 1843, where A is replaced by G; at the protein level this means replaces isoleucine at residue 615 with valine — a missense variant. Submitter rationale: Variant summary: CDH1 c.1843A>G (p.Ile615Val) results in a conservative amino acid change located in the Cadherin-like domain (IPR002126) of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251484 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1843A>G in individuals affected with Hereditary Diffuse Gastric Cancer and no experimental evidence demonstrating its impact on protein function have been reported. Four ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
