Uncertain significance for Congenital portosystemic shunt — the classification assigned by Department of Pediatrics, Graduate School of Medical Sciences, Kyushu University to NM_006667.5(PGRMC1):c.115A>G (p.Ile39Val), citing ACMG Guidelines, 2015: This missense variant was identified in a patient with congenital portosystemic shunt (CPSS) through family-based sequencing analysis. The variant is absent or extremely rare in population databases including gnomAD. In silico prediction tools, including CADD, suggest a deleterious effect on the protein. However, the association between this gene and CPSS has not been established. Therefore, the clinical significance of this variant is currently classified as a variant of uncertain significance (VUS).
