Pathogenic for Congenital adrenal hyperplasia due to cytochrome P450 oxidoreductase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001395413.1(POR):c.1841_1842del (p.Glu614fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Glu617Alafs*35) in the POR gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 64 amino acid(s) of the POR protein. This variant is present in population databases (no rsID available, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with POR-related conditions. This variant disrupts a region of the POR protein in which other variant(s) (p.His628Pro) have been determined to be pathogenic (PMID: 22162478). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
